La thiamine pyrophosphate (figure 1) est un coenzyme de transfert. Dérivé de la thiamine (vitamine B1), il est fixé à l'enzyme, c'est un groupement prosthétique.
Il participe aux réactions de décarboxylation.
La thiamine pyrophosphate est l'effecteur de la biosynthèse de la thiamine chez un certain nombre d'organismes pour lesquels ce n'est pas une vitamine (bactéries, plantes). L'expression des gènes correspondants est réprimée lorsque la concentration de thiamine pyrophosphate est suffisante. Ce contrôle passe par une régulation de la traduction des ARNm, qui implique un riboswitch.
