Lionel Loueke est né en Afrique de l'ouest au Bénin. Il s'installe en Côte d'Ivoire en 1990 pour étudier au National Institute of Art. Puis il reçoit une bourse pour étudier à Paris en France à l'American School of Modern Music de 1994 à 1998. En 1999, Lionel Loueke reçoit une bourse pour étudier à Berklee College of Music aux États-Unis à Boston, il termine et reçoit le diplôme un an après en 2000.

Lionel Loueke passe l'audition pour la Thelonious Monk Institute of Jazz à l'Université de Californie en 2001, et est sélectionné par un jury comprenant Herbie Hancock, Terence Blanchard et Wayne Shorter. il restera 2 ans à la Thelonious Monk Institute.

En 2002, pendant qu'il est toujours à la Thelonious Monk Institute, il commence à jouer avec le trompettiste Terence Blanchard. il enregistre 2 albums avec lui sur le label Blue Note.

Lionel Loueke fait partie du groupe Gilfema avec Ferenc Nemeth (batterie) et Massimo Biolcati (basse)
