La Protéine de Réplication A (ou RPA pour Replicative Protein A en anglais) est une protéine de liaison de la molécule d’ADN. Elle est impliquée dans plusieurs processus métaboliques chez les Eucaryotes, notamment dans la réplication, la réparation et la recombinaison de l’ADN.
Cette protéine fixe principalement l’ADN simple-brin. Elle peut également lier l’ADN double-brin, mais avec beaucoup moins d’affinité. La RPA est composée de trois sous-unité : la sous-unité 70 kDa, la 32 kDa et la sous-unité 14 kDa. C’est dans la sous-unité 70 kDa qu’est situé le site de liaison avec la molécule d’ADN.
