Il joue essentiellement de la contrebasse et de la basse électrique. Il est connu pour avoir un son très particulier, dû au fait qu'il joue sur des basses de marque Alembic. Il a démocratisé la technique du slap, créée à l'origine par Larry Graham, en l'appliquant au jazz. Il contribua grandement au nouveau rôle de la basse dans un groupe notamment dans les solos.

Biographie
Ses débuts
Après avoir été diplômé de l'école de musique de Philadelphie, il part à New York en 1971 et commence à jouer avec des personnalités du monde de la musique tels Horace Silver, Art Blakey, Dexter Gordon, Gato Barbieri, Joe Henderson, Pharoah Sanders, Gil Evans et Stan Getz.
Sa carrière
Il crée un groupe de jazz fusion, Return to Forever, avec le pianiste Chick Corea, avec lequel il sortira plusieurs albums. À la même période, il commence à faire des albums solo.
En 1991, Stanley Clarke reçoit un BMI Film & TV Awards pour le morceau Black on Black crime dans le film Boyz N the Hood.
Depuis 2008, il fait partie du groupe de bassistes SMV avec Marcus Miller et Victor Wooten.
