Manlia Scantilla était l'épouse du sénateur Marcus Didius Julianus. En 193, après l'assassinat de Pertinax par les prétoriens, c'est Didius Julianus qui leur acheta l'empire a cours d'une mise aux enchères qui scandalisa le peuple de Rome et les légions des frontières. Manlia Scantilla fut donc impératrice pendant quelques mois, jusqu'à ce que le Sénat, effrayé par l'arrivée de Septime Sévère qui menaçait de prendre Rome, se ravise et destitue Didius Julianus, qui fut assassiné peu après.
On ignore ce qu'il advint de Manlia Scantilla et de leur fille Didia Clara.
